Fabrication of colorimetric pH-indicator based on bioplastic support matrix and anthocyanins extracted from red cabbage

The present PhD thesis entitled’’ Colorimetric pH indicator films based amylose and anthocyanins’’ is submitted to the Department of Plant and Environmental Sciences, Faculty of Science, University of Copenhagen, to fulfill the requirements to obtain the degree of Doctor of Philosophy (Ph.D.) for the full time PhD programme. The experimental work was primarily carried out at Department of Plant and Environmental Sciences, Faculty of Science, University of Copenhagen under the guidance of the main supervisor Andreas Blennow. The entire project was conceived and realized in collaboration with Professor Mohammed Saddik Motawie from department of Plant and Environmental sciences. Moreover, associate professor Jacob Judas Kan Kirkensgaard, and Professor Rasmus Bro, and Post doc. Marta Bevilacqua at Department of food Science. In addition, Associate professor Heloisa N. Bordallo at Niels Bohr institute, Faculty of science Copenhagen University. The project was met a collaboration with the Teknologi Institute DMRI, where real experiments were supported by Tomas Jacobsen and a great team. The thesis consists of summaries in English and in Danish, the introduction about responsive packaging systems. The first article describes the first attempt of preparation and characterization of nanocomposite films based on amylose (AM) and cellulose nanofibers (CNF) derived from sugar beet pulp wastes. The second paper is a review provides a concise discussion on high amylose starch (HAS) applications and forms relevant to its functionality with emphasis on biomaterials and its interactions with other natural polysaccharides. The third manuscript demonstrates pH indicator films based on anthocyanins (RCA) immobilized in three different α-glucan matrices. Physical, mechanical and thermal properties, and color parameters have been tested. The fourth manuscript reports a pH sensitive composite film based on pure AM and CNF as carrier polymers, and pigments incorporated such as anthocyanins and neutral red. The pH indicator films were tested on minced meat and minced fish packed under modified atmosphere gas (MAP). The fifth manuscript describes a new route of synthesis between neutral red dye and maltooctaose oligosaccharides, to immobilize it in amylose matrix, and to reduce the neutral red toxicity.
